Area of Science:

  • Virology
  • Clinical Diagnostics
  • Molecular Biology

Background:

  • Accurate influenza virus detection is crucial for clinical diagnosis.
  • Low viral loads in samples can lead to false-negative results.
  • Existing transport media may not optimize viral detection sensitivity.

Purpose of the Study:

  • To develop and validate a modified transport medium (MTM) to improve influenza virus detection sensitivity.
  • To assess the impact of MTM on viral load and positive detection rates in clinical samples.
  • To compare the efficacy of MTM with other transport mediums for influenza diagnostics.

Main Methods:

  • Laboratory validation of MTM using stocked influenza A (IAV) and influenza B (IBV) viruses.
  • Testing MTM with clinical samples from 110 patients, collecting paired samples.
  • Quantification of viral load and positive detection rates using real-time polymerase chain reaction (RT-PCR) after MTM treatment.

Main Results:

  • MTM treatment increased detection sensitivity by 10-fold (H1N1, H9N2, IBV), 100-fold (H3N2), and 1,000-fold (H7N3) after 24h culturing.
  • Positive detection rates for IAV increased from 63.16% to 89.47% and for IBV from 36.36% to 72.73% after 72h culturing in MTM.
  • MTM demonstrated an 11.81% higher viral detection rate in clinical samples compared to other transport mediums (P=0.007).

Conclusions:

  • The modified transport medium (MTM) effectively increases influenza virus detection sensitivity.
  • MTM facilitates more accurate diagnosis of influenza infections by improving viral detection rates.
  • MTM shows significant clinical utility for enhancing influenza diagnostics.
