RNA-seq03:21

RNA-seq

9.9K
RNA sequencing, or RNA-Seq, is a high-throughput sequencing technology used to study the transcriptome of a cell. Transcriptomics helps to interpret the functional elements of a genome and identify the molecular constituents of an organism. Additionally, it also helps in understanding the development of an organism and the occurrence of diseases.
Before the discovery of RNA-seq, microarray-based methods and Sanger sequencing were used for transcriptome analysis. However, while...

Sanger Sequencing

754.0K
DNA sequencing is a fundamental technique that is routinely used in the biological sciences. This method can be applied to a range of questions at different scales - from the sequencing of a cloned DNA fragment or the study of a mutation in a gene up to whole-genome sequencing. However, despite the widespread use of sequencing today, it was not until 1977 that Fredrick Sanger and his collaborators developed the chain-termination method to decode DNA sequences. It relies on the separation of a...

Area of Science:

  • Virology
  • Genomics
  • Epidemiology

Background:

  • Usutu virus (USUV), a Flaviviridae family arbovirus, is emerging globally with documented spread across Africa, Europe, and Asia.
  • While often asymptomatic or causing mild illness, USUV can lead to severe neurological complications like encephalitis in humans.
  • The natural transmission cycle involves Culex mosquitoes and avian hosts, with humans and horses as incidental hosts.

Purpose of the Study:

  • To develop and validate a novel whole-genome amplicon-based sequencing approach for Usutu virus.
  • To enable robust genomic surveillance for understanding USUV evolution and transmission.

Main Methods:

  • Development of a whole-genome amplicon-based sequencing strategy.
  • Validation using Usutu virus strains from Senegal and Italy.
  • Testing the approach on samples from various vertebrate hosts.

Main Results:

  • The novel sequencing approach demonstrated good genome coverage.
  • The method proved effective across samples from different vertebrate hosts.
  • The study provides a valuable tool for Usutu virus genomic surveillance.

Conclusions:

  • A novel whole-genome sequencing method for Usutu virus has been successfully developed and validated.
  • This approach is suitable for genomic surveillance across diverse host samples.
  • The tool will aid in understanding Usutu virus genetic diversity, evolution, and transmission patterns.
