Genome-wide Association Studies-GWAS01:11

Genome-wide Association Studies-GWAS

13.2K
Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

8.7K
Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

Odds Ratio01:09

Odds Ratio

115
The odds ratio (OR) is a statistical measure used extensively in epidemiology and research to quantify the strength of association between exposure and outcome across different groups. Unlike relative risk, which compares the probabilities of an event occurring, the odds ratio compares the odds of an event occurring in the exposed group to the odds of it occurring in the unexposed group. The odds, in this context, are calculated as the probability of the event happening divided by the...

Randomized Experiments01:13

Randomized Experiments

6.8K
The randomization process involves assigning study participants randomly to experimental or control groups based on their probability of being equally assigned. Randomization is meant to eliminate selection bias and balance known and unknown confounding factors so that the control group is similar to the treatment group as much as possible. A computer program and a random number generator can be used to assign participants to groups in a way that minimizes bias.

Study Designs in Epidemiology01:20

Study Designs in Epidemiology

198
Epidemiological study designs are fundamental tools for investigating the distribution, determinants, and control of health conditions in populations. They help researchers understand the relationships between exposures and outcomes, and they broadly fall into two categories: "observational" and "experimental" studies.
Observational studies are those where the researcher does not intervene but rather observes natural variations. They include cross-sectional, cohort, and...

Summary

This study found no causal link between psoriasis or psoriatic arthritis and osteoarthritis (OA). While psoriasis can cause arthritis, it does not appear to directly cause OA development.

Area of Science:

  • Rheumatology and Genetics
  • Epidemiology

Background:

  • Psoriasis is associated with arthritis, particularly psoriatic arthritis.
  • A causal link between psoriasis, psoriatic arthritis, and osteoarthritis (OA) is not well-established.
  • Understanding this relationship is crucial for patient management and treatment.

Purpose of the Study:

  • To investigate the potential causal relationship between psoriasis and OA.
  • To examine the causal effect of psoriatic arthritis on different types of OA.
  • To differentiate between inflammatory arthritis and osteoarthritis in patients with psoriasis.

Main Methods:

  • Employed a 2-sample Mendelian randomization (MR) approach.
  • Utilized instrumental variables from large genome-wide association studies (GWAS) for psoriasis, psoriatic arthritis, and four OA types (HOA, KOA, TKR, THR).
  • Applied statistical methods including inverse variance weighting (IVW), MR-Egger, and weighted median estimation (WM).

Main Results:

  • Mendelian randomization analysis did not reveal a significant causal effect of psoriasis on hand OA (P=.381), knee OA (P=.725), total knee replacement (P=.488), or total hip replacement (P=.454).
  • Analysis also showed no evidence of a causal relationship between psoriatic arthritis and hand OA (P=.478), knee OA (P=.835), total hip replacement (P=.807), or total knee replacement (P=.860).
  • Statistical significance (P-values) and odds ratios (OR) for all tested relationships indicated a lack of causal association.

Conclusions:

  • This study provides no evidence supporting a causal link between psoriasis or psoriatic arthritis and the development of osteoarthritis.
  • The findings suggest that while psoriasis may be associated with inflammatory arthritis, it does not appear to be a direct cause of osteoarthritis.
  • Further research may be needed to explore indirect mechanisms or confounding factors in the association between psoriasis and OA.
