Background:

  • Multiple sclerosis (MS) is an autoimmune CNS disease.
  • Observational studies suggest a link between MS and male infertility.
  • The causal relationship between MS and sperm abnormalities remains unexamined.

Purpose of the Study:

  • To investigate the potential causal relationship between multiple sclerosis and abnormal spermatozoa using Mendelian randomization.
  • To assess if MS is a risk factor for sperm abnormalities.

Main Methods:

  • Utilized genome-wide association study data for single nucleotide polymorphisms (SNPs) as instrumental variables.
  • Employed two-sample Mendelian randomization (MR) with inverse variance weighting (IVW), MR-Egger, and MR-PRESSO.
  • Conducted sensitivity analyses including Cochran's Q, MR-Egger intercept, leave-one-out, and funnel plots.

Main Results:

  • A significant causal relationship was found between MS and abnormal sperm (OR 1.090, 95% CI [1.017-1.168], p = 0.014).
