Insights

Matrix metalloproteinase (MMP)-12 deficiency exacerbates kidney injury and inflammation in a mouse model of unilateral ureteric obstruction. MMP-12 appears to have protective roles in kidney disease by reducing inflammatory cytokines and macrophage accumulation.

Area of Science:

  • Nephrology
  • Immunology
  • Molecular Biology

Background:

  • Matrix metalloproteinase (MMP)-12 is known for immune regulation and anti-inflammatory effects.
  • The specific role of MMP-12 in kidney injury remains unclear.

Purpose of the Study:

  • To investigate the contribution of MMP-12 to tubulointerstitial injury in a mouse model of unilateral ureteric obstruction (UUO).

Main Methods:

  • Mice lacking MMP-12 (MMP-12-/-) and wild-type controls (MMP-12+/+) underwent UUO.
  • Chimeric mice with bone marrow deficient in MMP-12 were used to assess its role in monocytes/macrophages.
  • Bone marrow-derived macrophages (BMDMs) were isolated and stimulated with lipopolysaccharide.

Main Results:

  • MMP-12-/- mice exhibited worsened macrophage accumulation and interstitial fibrosis post-UUO.
  • Chimeric mice with MMP-12 deficient bone marrow also showed increased kidney injury.
  • UUO led to greater accumulation of Ly6C+ macrophages and elevated inflammatory cytokines (TNF-α, IL-1β, IL-6) in MMP-12-/- mice.

Conclusions:

  • MMP-12 appears to play a protective role in UUO-induced kidney injury.
  • MMP-12 mitigates kidney damage by reducing inflammatory cytokine production from macrophages and limiting macrophage accumulation.
