    Background:

    • Granulosa cells (GC) produce plasminogen activator (PA), stimulated by gonadotropins, suggesting a role in ovulation.
    • While LH is the primary ovulation-inducing hormone in rats, FSH shows greater potency in stimulating PA production by GC.

    Purpose of the Study:

    • To investigate the role of PA in the entire follicular wall during ovulation.
    • To determine the relative potencies of LH and FSH in stimulating follicular PA activity.
    • To explore the influence of steroids and arachidonic acid metabolites on LH-stimulated PA.

    Main Methods:

    • Assessed PA activity in intact ovarian follicles.
    • Administered serine protease inhibitors (epsilon-amino-caproic acid, benzamidine) to inhibit ovulation.
    • Cultured follicles with various hormones (LH, FSH, estradiol-17 beta, progesterone, testosterone) and inhibitors (aminoglutethimide phosphate, indomethacin, nordihydroguaiaretic acid).

    Main Results:

    • LH was 5-fold more potent than FSH in enhancing follicular PA activity in intact follicles.
    • Serine protease inhibitors significantly inhibited ovulation in a dose-dependent manner.
    • Estradiol-17 beta enhanced LH-stimulated PA, an effect reversed by specific inhibitors.
    • Arachidonic acid metabolites did not mediate LH-stimulated PA.

    Conclusions:

    • LH is a potent stimulator of follicular PA activity, playing a key role in ovulation.
    • Serine proteases, likely PA and plasmin, are confirmed to be involved in the ovulation process.
    • Estradiol-17 beta potentiates LH's effect on follicular PA, but arachidonic acid pathways are not involved.
