    Area of Science:

    • Cardiovascular Physiology
    • Biochemistry

    Background:

    • Ethanol's acute effects on cardiac protein synthesis are minimal.
    • Acetaldehyde, ethanol's metabolite, significantly reduces cardiac protein synthesis.
    • Chronic ethanol consumption during development may impair cardiac function.

    Purpose of the Study:

    • To investigate the effects of acute and chronic ethanol exposure on cardiac protein synthesis in guinea pigs.
    • To assess the impact of ethanol and its metabolite on cardiac contractile protein synthesis.
    • To evaluate the influence of chronic ethanol exposure on cardiac performance during ischemia.

    Main Methods:

    • In vitro assays of cardiac protein synthesis in guinea pig hearts.
    • Measurement of specific contractile protein synthesis (actin, myosin, tropomyosin).
    • Isolated working heart perfusion under normal and ischemic conditions.

    Main Results:

    • Acute ethanol did not alter protein synthesis; acetaldehyde decreased it.
    • Chronic ethanol exposure reduced right ventricular protein synthesis capacity, particularly actin.
    • Ethanol-exposed hearts showed impaired pressure maintenance and increased resistance during ischemia.

    Conclusions:

    • Acetaldehyde acutely inhibits cardiac protein synthesis.
    • Prolonged ethanol exposure during maturation impairs cardiac protein synthesis and function.
    • Ethanol may compromise the heart's ability to handle ischemic stress due to altered protein metabolism.
