Three new pregnane steroids were isolated from Marsdenia tenacissima roots. One compound, 11α-nicotinoyl-17β-marsdenin, showed significant cytotoxic activity against lung and ovarian cancer cells.
Area of Science:
- Natural Product Chemistry
- Medicinal Chemistry
- Pharmacology
Background:
- Marsdenia tenacissima is a plant source of bioactive compounds.
- Pregnane steroids represent a class of natural products with diverse biological activities.
Purpose of the Study:
- To isolate and characterize new pregnane steroids from Marsdenia tenacissima.
- To evaluate the cytotoxic activities of isolated compounds against human cancer cell lines.
Main Methods:
- Isolation and purification of compounds using chromatographic techniques.
- Structure elucidation via NMR spectroscopy, mass spectrometry, and X-ray diffraction.
- Cytotoxicity assays using A549, SKOV-3, MGC 803, and MCF-7 cancer cell lines.
Main Results:
- Three new pregnane steroids (1-3) and two known analogues (4-5) were identified.
- Compound 3 (11α-nicotinoyl-17β-marsdenin) demonstrated significant cytotoxic effects against A549 and SKOV-3 cells (IC50 values of 16.79 μM and 12.30 μM, respectively).
- Compounds showed moderate cytotoxicity against MGC803 and MCF-7 cells.
Conclusions:
- Marsdenia tenacissima is a valuable source of structurally novel pregnane steroids.
- 11α-nicotinoyl-17β-marsdenin possesses potent cytotoxic properties against specific human cancer types, warranting further investigation.
- The study contributes to the understanding of natural products' potential in cancer therapy.
