    Background:

    • Insertion sequences (IS) are mobile genetic elements that can cause mutations.
    • The IS4 element in E. coli has a known integration site.
    • Gene duplications can arise from various DNA repair or recombination mechanisms.

    Purpose of the Study:

    • To characterize the molecular mechanism of IS4 integration into the galT gene.
    • To determine the nature of DNA sequences duplicated adjacent to IS4 insertions.
    • To investigate potential relationships between IS4 integration and gene duplication events.

    Main Methods:

    • DNA sequencing (Sanger sequencing) was used to analyze the junctions between IS4 and the galT gene.
    • Cloning of the IS4 element from the E. coli K12 chromosome.
