    Barbiturates interfere with adenosine transport in the brain. These drugs inhibit adenosine uptake and enhance its release, potentially explaining their depressant effects.

    Purpose of the Study:

    • Synaptosomes from rodent brains were used to study adenosine uptake and release.
    • [3H]adenosine was employed to track transport dynamics.
    • Various barbiturates (pentobarbital, methohexital, phenobarbital, CHEB) were tested.

    Main Results:

    • All tested barbiturates inhibited [3H]adenosine uptake.
    • Pentobarbital increased potassium-induced release of [3H]adenosine.
    • Barbiturates did not alter adenosine metabolite distribution.

    Conclusions:

    • Barbiturates may exert depressant effects by disrupting adenosine homeostasis.
    • Inhibition of adenosine reuptake and enhanced release contribute to elevated synaptic adenosine levels.
