Obesity increases cardiovascular disease risk. This study found losartan and perindopril reduced pro-inflammatory IL-6 but increased ACE2 levels in obese rat adipose tissue.
Area of Science:
- Endocrinology
- Cardiovascular Research
- Pharmacology
Background:
- Obesity is a global health issue linked to cardiovascular diseases.
- Adipocytes express angiotensin-converting enzyme 2 receptors (ACE2) and adipocytokines, potentially triggering cytokine storms.
- Elevated interleukin-6 (IL-6) is associated with inflammation and cardiovascular complications.
Purpose of the Study:
- To investigate the effects of perindopril and losartan on ACE2 and IL-6 levels in adipose tissue of obese rats.
- To assess the potential of these drugs in modulating inflammation and ACE2 expression in obesity-related cardiovascular risk.
Main Methods:
- An in vivo true experimental design with a post-test-only control group was employed.
- Twenty-four male albino rats were divided into non-obese (control), obese (untreated), obese treated with perindopril, and obese treated with losartan groups.
- ACE2 and IL-6 levels in visceral fat tissue were quantified using enzyme-linked immunosorbent assay (ELISA) after 4 weeks of treatment.
Main Results:
- Losartan significantly increased ACE2 levels in obese rats compared to perindopril and untreated obese controls (p=0.011 and p=0.004, respectively).
- Both perindopril and losartan significantly reduced IL-6 levels in obese rats compared to the untreated obese group (p=0.020 and p=0.002, respectively).
Conclusions:
- Perindopril and losartan demonstrate potential in managing obesity-related cardiovascular risks.
- These drugs may suppress pro-inflammatory markers like IL-6 while enhancing protective ACE2 levels in adipose tissue.
