    Human monocytes express HLA-DR antigens, which decrease in vitro without lymphocytes or interferon-gamma (IFN-γ). Lymphocytes and low doses of IFN-γ maintain monocyte DR antigen expression, suggesting lymphocytes produce IFN-γ.

    Background:

    • Human monocytes express Human Leukocyte Antigen - antigen D Related (HLA-DR) antigens.
    • The regulation of HLA-DR expression on monocytes during in vitro cultivation is not fully understood.

    Purpose of the Study:

    • To investigate the kinetics of HLA-DR antigen expression in human peripheral blood monocytes during in vitro cultivation.
    • To determine the role of lymphocytes and interferon-gamma (IFN-γ) in maintaining monocyte HLA-DR expression.

    Main Methods:

    • Monocytes were isolated by glass adherence and cultured in vitro.
    • Indirect fluorescence staining with a monoclonal antibody (BL-DR/1) was used to assess DR antigen expression.
    • The effects of lymphocyte depletion, addition of lymphocytes, and exogenous IFN-γ were evaluated.

    • Approximately 60% of freshly isolated monocytes expressed DR antigens.
    • Monocytes depleted of lymphocytes gradually lost DR antigen expression during cultivation.
    • Addition of lymphocytes or low doses of IFN-γ (≥1 IU/ml) prevented the decrease in DR antigen expression.
    • Cyclosporin A experiments suggested contaminating lymphocytes spontaneously produce sufficient IFN-γ to maintain DR antigens on monocytes.

    Conclusions:

    • Monocyte HLA-DR antigen expression is dependent on the presence of lymphocytes or exogenous IFN-γ.
    • Lymphocytes play a crucial role in maintaining monocyte HLA-DR expression, likely through IFN-γ production.
    • IFN-γ is a potent inducer of HLA-DR expression on cultured monocytes.
