Area of Science:

  • * Biochemistry and Proteomics
  • * Food Science and Nutrition

Background:

  • * Bovine colostrum is vital for neonatal nutrition, immunity, and growth, containing bioactive compounds like peptides.
  • * Endogenous peptides (EPs) are of significant scientific and commercial interest due to their diverse bioactivities (e.g., antimicrobial, anti-inflammatory).
  • * Peptidomics research focuses on identifying and characterizing peptides from biological sources.

Purpose of the Study:

  • * To systematically compare four distinct peptide extraction methods for low molecular weight EPs from bovine colostrum.
  • * To identify the most effective extraction technique for maximizing EP identification in colostrum.
  • * To pioneer the comparative analysis of EP extraction methods specifically for colostrum.

Main Methods:

  • * Extraction of low molecular weight endogenous peptides (EPs) from bovine colostrum.
  • * Systematic comparison of four different peptide extraction techniques.
  • * Utilized trichloroacetic acid (TCA) and molecular weight cut off (MWCO) methods.

Main Results:

  • * Successfully extracted over 3200 endogenous peptides (EPs) from bovine colostrum.
  • * The trichloroacetic acid (TCA) and molecular weight cut off (MWCO) methods demonstrated superior efficiency.
  • * Identified EPs with predicted bioactivities using in silico tools.

Conclusions:

  • * The study successfully established an effective method for extracting a large number of EPs from bovine colostrum.
  • * Trichloroacetic acid (TCA) and molecular weight cut off (MWCO) are recommended for colostrum EP analysis.
  • * The identified EPs hold potential for various health-related applications.
