    Background:

    • The aging process can affect gastrointestinal function.
    • Understanding changes in the small intestine during aging is crucial for explaining functional alterations.

    Purpose of the Study:

    • To investigate the impact of aging on the distribution, content, and concentration of epithelial cell enzymes in the small intestine.
    • To compare enzyme profiles in aging rats with those in young adult rats.

    Main Methods:

    • Enzyme activities (sucrase, maltase, lactase, adenosine deaminase, thymidine kinase) were measured in mucosal homogenates and specific cells along the crypt-villus axis.
    • Cryostat sectioning was used to isolate cells from different levels of the crypt-villus column.
    • Fisher 344 rats (aging and young adult) were used for the study.

    Main Results:

    • Specific activities of sucrase, maltase, lactase, and adenosine deaminase were lower in the upper intestines of aging rats compared to young rats.
    • Thymidine kinase specific activity and content were higher in aging rats.
    • The typical spatial pattern of enzyme expression along the crypt-villus axis was maintained in aging rats, but initiation and duration of expression were delayed.
    • Reduced enzyme activities in aging animals were attributed to an increased proportion of undifferentiated villus epithelial cells.

    Conclusions:

    • Aging alters the expression patterns of small intestinal epithelial cell enzymes.
    • Functional changes in the aging intestine can occur without significant structural modifications.
    • The findings help explain altered intestinal function during aging.
