    Background:

    • The hypothalamus-pituitary-testis (HPT) axis regulates male reproductive function.
    • Understanding the intricate functional interrelations within the HPT axis requires precise experimental models.
    • Previous methods may lack the sensitivity or specificity to fully elucidate these complex interactions.

    Purpose of the Study:

    • To establish and validate a novel in vitro system for studying the HPT axis.
    • To assess the sensitivity and specificity of this system in measuring key hormonal responses.
    • To provide a tool for evaluating the effects of external agents on HPT axis function.

    Main Methods:

    • An in vitro system was created using rat hypothalamic tissue, isolated anterior pituitary cells, and Leydig cells.
    • Testosterone production by Leydig cells was quantified as the primary endpoint.
    • Sensitivity thresholds for human chorionic gonadotropin (hCG) on Leydig cells and gonadotropin-releasing hormone (GnRH) on pituitary cells were determined.

    • The system offers a sensitive and specific method for endocrine research and drug evaluation.
