This study developed novel solid lipid nanoparticles (SLNs) to enhance caffeine (CA) delivery for cellulite treatment. The resulting CA-SLN-Ngel formulation effectively reduced subcutaneous fat tissue, offering a promising topical solution for cellulite.
Area of Science:
- Nanotechnology
- Dermatology
- Cosmeceuticals
Background:
- Cellulite (CLT) is a common cosmetic concern affecting post-adolescent women.
- Current treatments like liposuction and mesotherapy are invasive and risky.
- Topical treatments are preferred, but caffeine's (CA) hydrophilicity limits skin penetration.
Purpose of the Study:
- To develop solid lipid nanoparticles (SLNs) for enhanced topical delivery of caffeine (CA).
- To evaluate the anti-cellulite efficacy of CA-loaded SLNs incorporated into a hydrogel formulation (CA-SLN-Ngel).
Main Methods:
- Caffeine was loaded into SLNs using high shear homogenization and ultrasonication.
- Formulation variables were optimized for entrapment efficiency, particle size, zeta potential, and polydispersity index.
- CA-SLN-Ngel was prepared and its physical properties, in vitro release, and histological effects on subcutaneous fat tissue were assessed.
Main Results:
- Optimized CA-SLNs (CA-SLN-4) achieved high entrapment efficiency (92.03%), small particle size (232.7 nm), and optimal zeta potential (-25.15 mV).
- CA-SLN-4 exhibited superior caffeine release (99.44% at 1h) and demonstrated successful transepidermal permeation.
- CA-SLN-Ngel significantly reduced subcutaneous fat tissue thickness compared to caffeine gel and untreated groups.
Conclusions:
- Solid lipid nanoparticles effectively enhance caffeine's skin delivery and anti-cellulite effects.
- CA-SLN-Ngel represents a promising nanocosmeceutical gel for topical cellulite treatment.
- The study highlights the importance of skin appendages for effective topical drug delivery.
