Area of Science:

  • Ophthalmology
  • Neuroscience
  • Genomics

Background:

  • Myopia, a global vision impairment, is linked to retinal cellular changes.
  • Lens-induced myopia (LIM) models are crucial for understanding myopia progression.
  • Electroacupuncture (EA) shows potential therapeutic benefits for experimental myopia.

Purpose of the Study:

  • To create a comprehensive single-cell atlas of retinal cells in LIM and EA-treated guinea pigs.
  • To investigate the molecular mechanisms underlying EA's effect on myopia progression.
  • To explore EA's role in enhancing retinal mitochondrial function to combat myopia.

Main Methods:

  • Single-cell RNA sequencing (scRNA-seq) was employed to analyze gene expression at the individual cell level.
  • Retinal tissues from guinea pigs with LIM and those receiving EA intervention were profiled.
  • Mitochondrial function markers were specifically examined in relation to myopia.

Main Results:

  • scRNA-seq identified ten distinct cell clusters within the retina.
  • Mitochondrial respiratory chain genes (COX3, ND4, ND2) in Cones and Muller glia cells were significantly associated with LIM.
  • EA intervention demonstrated a positive effect on improving retinal mitochondrial function.

Conclusions:

  • A detailed single-cell atlas of the retina in LIM and EA-treated guinea pigs was successfully established.
  • EA treatment was found to improve retinal mitochondrial function.
  • These findings suggest EA is a promising intervention for slowing experimental myopia progression in guinea pigs.
