    Background:

    • Traditional methods for measuring left ventricular volumes and dimensions in rats can be challenging due to their small size.

    Purpose of the Study:

    • To evaluate the accuracy and feasibility of using high-speed biplane ventriculography for quantifying rat left ventricular (LV) dimensions and volumes.
    • To establish reference values for LV parameters in healthy anesthetized rats.

    Main Methods:

    • Biplane ventriculography was performed on nine anesthetized rats at high frame rates (500 frames/s) with direct enlargement.
    • Left ventricular silhouettes were digitized to calculate end-diastolic volume, end-systolic volume, stroke volume, ejection fraction, and cardiac output.
    • Methyl methacrylate casts of 11 rat left ventricles were used to validate volume measurements against water displacement and monoplane/biplane methods.

    • Accurate LV dimensions and volumes were obtained, including end-diastolic volume (0.60 ± 0.03 ml) and ejection fraction (0.63 ± 0.02).
    • Cardiac output was measured at 118 ± 7 ml/min.
    • High correlation (r = 0.99 ± 0.02) was found between cast volumes and ventriculography measurements, confirming method accuracy.

    Conclusions:

    • High-speed biplane ventriculography is a validated and accurate method for assessing rat left ventricular function.
    • This technique provides reliable volumetric and dimensional data essential for cardiovascular research in rodent models.
