Evolutionary Relationships through Genome Comparisons

5.7K
Genome comparison is one of the excellent ways to interpret the evolutionary relationships between organisms. The basic principle of genome comparison is that if two species share a common feature, it is likely encoded by the DNA sequence conserved between both species. The advent of genome sequencing technologies in the late 20th century enabled scientists to understand the concept of conservation of domains between species and helped them to deduce evolutionary relationships across diverse...

Area of Science:

  • Microbiology
  • Genomics
  • Taxonomy

Background:

  • The *Providencia* genus is gaining clinical importance due to antibiotic resistance.
  • Genomic and taxonomic understanding of *Providencia* is currently being revised.
  • Multidrug resistance in *Providencia* species necessitates updated characterization.

Purpose of the Study:

  • To outline and characterize the *P. stuartii* species using genomic approaches.
  • To establish robust genomic definitions for *P. stuartii*.
  • To develop novel typing schemes for *P. stuartii*.

Main Methods:

  • Retrieved *Providencia* genomes from GenBank.
  • Performed genome-based species definition using average nucleotide identity (ANI) and alignment-free methods.
  • Devised MultiLocus Sequence Typing (MLST) and core-genome MLST (cgMLST) schemes.

Main Results:

  • Defined genomes reliably identifiable as *P. stuartii*.
  • Developed a 7-locus MLST scheme.
  • Developed a 2,296-locus cgMLST scheme.

Conclusions:

  • This work provides a framework for understanding *P. stuartii*.
  • *P. stuartii* should be recognized as an emerging multidrug-resistant pathogen.
  • The developed typing schemes will aid in tracking *Providencia* infections.
