    Summary

    Respiratory syncytial virus (RSV) activates complement pathways in infected human cells. This complement activation, along with antibody-dependent cell lysis, may contribute to RSV disease pathogenesis.

    Background:

    • Respiratory syncytial virus (RSV) is a significant pediatric respiratory pathogen.
    • The role of complement (C) in respiratory secretions during RSV infection is not well understood.
    • RSV-infected respiratory epithelial cells express viral surface antigens.

    Purpose of the Study:

    • To investigate the ability of RSV-infected human cells to activate the complement system.
    • To determine the involvement of complement pathways in the cytolysis of RSV-infected cells.

    Main Methods:

    • RSV-infected HeLa cells were used to study complement activation.
    • Classical and alternative complement pathways activation was measured by C3 cleavage.
    • Cytolysis of RSV-infected cells was assessed using a chromium release assay.

    Main Results:

    • RSV-infected human cells activated both classical and alternative complement pathways.
    • Complement activation was enhanced in the presence of antibodies.
    • Both complement pathways, with antibody presence, contributed to the lysis of RSV-infected cells.
