Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. Summary

Rab44 deficiency reduces inflammation and bone destruction in a mouse model of rheumatoid arthritis (RA). This study shows Rab44-knockout mice have less joint inflammation and bone loss in collagen antibody-induced arthritis (CAIA).

Background:

  • Rheumatoid arthritis (RA) involves joint inflammation and bone destruction driven by immune cells and osteoclasts.
  • Collagen antibody-induced arthritis (CAIA) is a key mouse model for studying RA pathogenesis.
  • The novel gene Rab44, highly expressed in immune cells and osteoclasts, was previously identified.

Purpose of the Study:

  • To investigate the role of Rab44 in inflammation, cell infiltration, and bone destruction.
  • To assess the impact of Rab44 deficiency on the CAIA mouse model.

Main Methods:

  • Induction of CAIA in Rab44-knockout (KO) and wild-type (WT) mice.
  • Analysis of inflammatory markers, cell infiltration, and bone destruction in the joints.
  • Quantification of mRNA levels for arthritis-related genes.

Main Results:

  • Rab44-KO mice exhibited significantly reduced joint inflammation and cell infiltration compared to WT mice.
  • Decreased mRNA expression of inflammation, cartilage turnover, bone formation, and bone resorption markers was observed in Rab44-KO CAIA mice.
  • Rab44 deficiency led to predominant M2 macrophage infiltration and reduced bone loss in CAIA mice.

Conclusions:

  • Rab44 deficiency ameliorates arthritis progression in the CAIA mouse model.
  • Rab44 plays a critical role in immune cell-mediated joint inflammation and bone destruction in RA.
