Interleukin-33 (IL-33) negatively impacts adipogenesis, while its receptor, suppression of tumorigenicity 2 (ST2), promotes it. The IL-33/ST2 axis regulates adipogenesis via the TRAF6/RelA pathway, offering new obesity prevention strategies.
Area of Science:
- Cell Biology
- Endocrinology
- Molecular Biology
Background:
- Adipogenesis regulation is crucial for obesity prevention.
- Interleukin-33 (IL-33) and its receptor, suppression of tumorigenicity 2 (ST2), are implicated in adipogenesis.
- Understanding their precise roles and mechanisms is essential.
Purpose of the Study:
- To investigate the function and regulatory mechanism of the IL-33/ST2 axis in adipogenesis.
- To elucidate the molecular pathways involved in IL-33/ST2-mediated regulation of preadipocyte differentiation.
Main Methods:
- Oil Red O staining for lipid droplet accumulation.
- Quantitative real-time PCR (qRT-PCR) and Western blotting for gene and protein expression analysis.
- Gain- and loss-of-function experiments in 3T3-L1 preadipocytes.
Main Results:
- IL-33 negatively correlates with adipogenesis, while ST2 (both ST2L and sST2) positively correlates in 3T3-L1 cells.
- The IL-33/ST2 axis modulates adipogenesis by regulating the TRAF6/RelA pathway.
- Downregulating ST2 inhibits the IL-33/ST2 axis, reducing TRAF6 and RelA expression, thereby suppressing adipogenesis.
Conclusions:
- The IL-33/ST2 axis plays a significant role in regulating adipogenesis through the TRAF6/RelA pathway.
- This axis represents a novel mechanism controlling preadipocyte differentiation.
- Targeting the IL-33/ST2 pathway offers potential therapeutic strategies for obesity prevention.
