Area of Science:

  • Pulmonary Medicine
  • Genomics
  • Transcriptomics
  • Molecular Biology

Background:

  • Acute Interstitial Pneumonia (AIP) is a severe, poorly understood lung disease within the idiopathic interstitial pneumonia spectrum.
  • Limited knowledge of AIP's molecular basis hinders the identification of effective therapeutic strategies.

Purpose of the Study:

  • To elucidate the genomic and transcriptomic profiles of AIP.
  • To uncover the underlying pathophysiology of AIP.
  • To identify potential therapeutic targets for AIP.

Main Methods:

  • Comprehensive genomic and transcriptomic analysis of lung tissue from 15 AIP patients.
  • Assessment of differentially expressed genes (DEGs), exonic coding variants, and expression quantitative trait loci (eQTL) profiles.
  • Comparison of molecular patterns with idiopathic pulmonary fibrosis (IPF) and other interstitial lung diseases (ILDs).

Main Results:

  • Significant upregulation of Type I interferon receptor and keratin filament genes; downregulation of focal adhesion and endothelial integrity genes in AIP.
  • Distinct molecular signature of AIP compared to IPF and other ILDs.
  • Identification of mutations in keratin and extracellular matrix genes.
  • eQTL profiling provided insights into genetic regulation of gene expression in AIP.

Conclusions:

  • AIP possesses a unique molecular landscape, distinct from other ILDs.
  • Findings provide a foundation for developing novel diagnostic markers and therapeutic interventions for AIP.
  • Genomic and transcriptomic insights pave the way for targeted treatments for this severe lung condition.
