Background:

  • Serotonin (5-HT) plays a role in stress response, partly mediated by 5-HT1a and 5-HT7 receptor subtypes.
  • While 5-HT1a receptor activation aids adaptation to stress, the role of 5-HT7 receptors in the hippocampus requires further investigation.
  • Hippocampal 5-HT7 receptor (5HT7r) mRNA expression increases after stress, but its functional significance in stress response is unclear.

Purpose of the Study:

  • To investigate if blocking hippocampal 5HT7r can prevent or reduce the behavioral effects of stress.
  • To determine the role of hippocampal 5HT7r in the consolidation and retrieval of stress-related memories.

Main Methods:

  • Adult male Wistar rats received bilateral hippocampal cannulations.
  • Rats were exposed to restraint stress (2h) or forced swim stress (15min).
  • SB 258741, a selective 5HT7r antagonist, was administered intra-hippocampally before stress, after stress, or before testing.

Main Results:

  • Hippocampal 5HT7r blockade administered immediately after stress exposure attenuated stress-induced behavioral changes in both restraint and forced swim tests.
  • Administration of the antagonist before testing also reduced behavioral effects in previously stressed rats.
  • Blocking 5HT7r before stress did not prevent behavioral changes, indicating a role in memory consolidation or retrieval rather than acute stress response.

Conclusions:

  • Activation of hippocampal 5-HT7 receptors is critical for the consolidation and retrieval of aversive memories associated with stressful experiences.
  • Stress-induced changes in 5HT7r expression likely mediate these effects.
  • Targeting hippocampal 5HT7r may offer therapeutic potential for stress-related disorders.
