Hydrochlorothiazide lowers urinary hydroxyproline in parathyroidectomized rats.

    Summary

Hydrochlorothiazide treatment significantly reduced urinary hydroxyproline excretion in parathyroidectomized rats. This effect occurred independently of changes in parathyroid hormone-mediated bone turnover.

    Area of Science:

    • Biochemistry
    • Pharmacology
    • Nephrology

    Background:

    • Urinary hydroxyproline is a marker of bone collagen breakdown.
    • Thiazide diuretics are commonly used for hypertension and edema.
    • The effect of thiazides on bone metabolism is not fully understood.

    Purpose of the Study:

    • To investigate the impact of hydrochlorothiazide on urinary hydroxyproline excretion.
    • To determine if this effect is linked to parathyroid hormone activity.

    Main Methods:

    • Parathyroidectomy was performed on rats.
    • Rats were treated with hydrochlorothiazide or a placebo.
    • Urinary hydroxyproline excretion and glomerular filtration rate were measured.

    Main Results:

    Related Experiment Videos

    • Hydrochlorothiazide treatment led to a significant decrease in urinary hydroxyproline excretion.
    • This reduction occurred without a significant change in glomerular filtration rate.
    • The findings suggest a direct effect of hydrochlorothiazide on collagen metabolism.

    Conclusions:

    • Hydrochlorothiazide effectively reduces urinary hydroxyproline excretion.
    • This effect is independent of parathyroid hormone-mediated bone turnover.
    • Thiazide diuretics may have a direct impact on collagen metabolism in bone.
