Amyloid Fibrils03:03

Amyloid Fibrils

9.2K
Amyloid fibrils are aggregates of misfolded proteins.  Under most circumstances, misfolded proteins are either refolded by chaperone proteins or degraded by the proteasome. However, in the case of a mutation or a disease, these proteins can accumulate to form large clusters and often further assemble to form elongated fibers, called fibrils.
Amyloid deposits were observed as early as 1639 in the liver and the spleen.   In 1854, Rudolph Virchow performed iodine staining,...

Alzheimer's Disease: Overview

444
Alzheimer's Disease (AD) is a continually advancing neurodegenerative disorder, distinguished by escalating memory loss, cognitive dysfunction, and dementia. The disease unfolds in three stages: preclinical, mild cognitive impairment (MCI), and dementia. Its onset is insidious, and the progression gradual, with the cause not well explained by other disorders.
The clinical diagnosis of AD hinges on the presence of memory and other cognitive impairments. Biomarkers, such as changes in Aβ...

Area of Science:

  • Cardiology
  • Hematology
  • Pathology

Background:

  • Amyloids are protein fibrils implicated in localized or systemic diseases.
  • Systemic amyloidosis frequently affects the heart, kidneys, liver, and nervous system.
  • Immunoglobulin light chain (AL) and transthyretin (TTR) amyloidosis are the most common cardiac types.

Purpose of the Study:

  • To review cardiac manifestations of amyloidosis.
  • To propose a diagnostic pathway for cardiac amyloidosis.
  • To highlight advancements in amyloidosis treatment.

Main Methods:

  • Literature review of cardiac amyloidosis features.
  • Analysis of diagnostic "red flags" (clinical, imaging).
  • Summary of current therapeutic strategies.

Main Results:

  • Cardiac amyloidosis presents with non-specific heart failure symptoms.
  • Specific clinical and imaging findings can indicate amyloidosis.
  • Therapeutic options for amyloidosis are rapidly expanding.

Conclusions:

  • Early identification of cardiac amyloidosis is vital.
  • A simplified diagnostic approach can aid clinicians.
  • The evolving treatment landscape offers new hope for patients.
