New influenza A virus (IAV) vaccines using virus-like particles (VLPs) show promise for broader protection and faster production. These VLP vaccines may overcome limitations of current flu shots, offering improved immunity.
Area of Science:
- Virology and Immunology
- Vaccine Technology Development
Background:
- Annual influenza A virus (IAV) infections cause significant mortality and morbidity globally.
- Current egg-based flu vaccines have limitations including production time, cost, moderate efficacy in the elderly, and lack of cross-protection.
- Need for improved vaccine strategies to address conserved IAV antigens and enhance immunogenicity.
Purpose of the Study:
- To review recent advancements in broadly effective IAV antigens and their display on virus-like particles (VLPs).
- To discuss current IAV VLP vaccine production systems and administration routes.
- To explore the potential of VLPs in overcoming current vaccine limitations and advancing intranasal vaccination.
Main Methods:
- Review of pre-clinical and clinical studies on IAV VLP vaccines.
- Analysis of strategies for displaying conserved IAV antigens on VLPs.
- Examination of VLP vaccine production systems and administration methods (intranasal, transdermal).
Main Results:
- VLPs can display multiple conserved IAV antigens, enhancing immune responses.
- Various production systems for IAV VLP vaccines are available and under development.
- Intranasal and transdermal administration routes show potential for VLP vaccines.
Conclusions:
- IAV VLP vaccines represent a promising strategy for developing broadly protective and rapidly produced influenza vaccines.
- VLPs offer advantages in overcoming immunological limitations and enabling novel administration routes like intranasal vaccination.
- Further clinical trials and expedited production are anticipated for the commercialization of IAV VLP vaccines.
