Circulating precursor-miR-488 (pre-miR-488) and miR-488-5p are elevated in gastric cancer patients' blood and indicate poor prognosis. These microRNAs may serve as novel biomarkers for predicting patient outcomes.
Area of Science:
- Oncology
- Molecular Biology
- Biomarker Discovery
Background:
- MicroRNAs (miRNAs) are emerging as non-invasive biomarkers in liquid biopsies.
- Gastric cancer (GC) diagnosis and prognosis can be improved by identifying novel biomarkers.
Purpose of the Study:
- To investigate the clinical significance of circulating precursor-miR-488 (pre-miR-488) and miR-488-5p in gastric cancer patients.
- To evaluate their potential as prognostic biomarkers in GC.
Main Methods:
- Quantitative reverse transcription PCR (RT-qPCR) to measure pre-miR-488 and miR-488-5p expression in tumor tissues and blood.
- Gene Set Enrichment Analysis (GSEA) on TCGA dataset for pathway analysis.
- Exosomal pre-miR-488 detection in patient plasma.
Main Results:
- Both pre-miR-488 and miR-488-5p were downregulated in GC tumor tissues but upregulated in the blood of GC patients compared to healthy controls.
- Low blood expression of pre-miR-488 or miR-488-5p correlated with poor prognosis in GC patients.
- Low pre-miR-488 blood expression was an independent predictor of poor overall survival; miR-488-5p showed negative correlation with epithelial-mesenchymal transition and hypoxia pathways. Pre-miR-488 was found in exosomes.
Conclusions:
- Circulating pre-miR-488 and miR-488-5p show potential as novel prognostic biomarkers for gastric cancer.
- These miRNAs could aid in predicting clinical outcomes and guiding therapeutic strategies for GC patients.
