MicroRNAs

3.0K
MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

Area of Science:

  • Molecular Biology
  • Virology
  • Genetics

Background:

  • Coronavirus disease 2019 (COVID-19) presents diverse clinical symptoms.
  • Host microRNAs (miRNAs) can influence COVID-19 progression and complications.
  • Understanding host factors like miRNA expression is crucial for managing severe SARS-CoV-2 infection.

Purpose of the Study:

  • To investigate the expression patterns of endogenous miRNAs in severe COVID-19 patients.
  • To compare miRNA expression between 80 severe COVID-19 cases and healthy controls.
  • To identify specific miRNAs associated with severe SARS-CoV-2 infection.

Main Methods:

  • miRNA expression screening using TaqMan Low-Density Array.
  • Validation of specific miRNA expression (miR-490-3p, miR-195-5p, miR-454-3p, miR-431-5p) via RT-qPCR.
  • In silico analysis for target identification and pathway prediction.

Main Results:

  • Four miRNAs (miR-490-3p, miR-195-5p, miR-454-3p, miR-431-5p) were significantly over-expressed in severe COVID-19 patients compared to controls.
  • miR-490-3p showed over-expression in both male and female COVID-19 patients.
  • These findings highlight specific miRNA dysregulation in severe SARS-CoV-2 infection.

Conclusions:

  • Specific miRNAs demonstrate altered expression in severe COVID-19.
  • These over-expressed miRNAs may function as potential biomarkers for predicting COVID-19 clinical outcomes.
  • Further research into miRNA roles could inform therapeutic strategies for severe coronavirus disease 2019.
