Area of Science:

  • Neuroscience
  • Developmental Biology
  • Genetics

Background:

  • Neuronal diversity in Drosophila arises from asymmetric division of ~100 neural stem cells.
  • Accurate single-cell resolution lineage mapping is vital for understanding neuronal specification.
  • Current lineage tracing methods are time-consuming and labor-intensive.

Purpose of the Study:

  • To introduce CLADES (cell lineage access driven by an edition sequence) as an improved method for neuronal lineage tracing.
  • To address the limitations of existing high-resolution lineage tracing techniques.
  • To provide best practices for utilizing CLADES.

Main Methods:

  • CLADES employs a genetic cascade of reporter activations and deactivations.
  • Color-coded markers delineate lineage progression.
  • The system is designed for efficient tracing with approximately 100 samples.

Main Results:

  • CLADES effectively traces the birth order of neurons.
  • The method facilitates detailed mapping of neuronal lineages.
  • The technique demonstrates efficiency and overcomes previous limitations.

Conclusions:

  • CLADES offers a revolutionary approach to neuronal lineage tracing in Drosophila.
  • The system provides a time- and labor-efficient solution for high-resolution lineage mapping.
  • CLADES has potential applications in tracking biological events and cell-type production for therapeutics.
