Mesenchymal Stem Cells01:19

Mesenchymal Stem Cells

4.6K
Mesenchymal stem cells (MSCs) are adult stem cells that can differentiate into most connective tissue cell types, except for hematopoietic cells, depending upon the source of MSCs. For example, bone-marrow-derived MSCs (BM-MSCs) can differentiate into osteocytes, hepatocytes, and pancreatic and neuronal cells. MSCs can be isolated from various sources such as bone marrow, placenta, adipose tissue, teeth, and Wharton’s jelly, a gelatinous substance in the umbilical cord. The ease of their...

Area of Science:

  • Regenerative Medicine
  • Stem Cell Biology
  • Molecular Biology

Background:

  • Tonsil-derived mesenchymal stem cells (TMSCs) show promise in regenerative medicine due to their proliferative and differentiation capabilities.
  • Optimizing TMSC yield and differentiation potential requires understanding culture conditions.

Purpose of the Study:

  • To investigate the impact of serial subculture on TMSC gene expression.
  • To identify optimal passage numbers for maintaining TMSC differentiation capacity.

Main Methods:

  • Gene expression analysis of TMSCs at passages 2, 4, 6, and 8.
  • Enrichment analysis using KEGG and Reactome pathways to identify significant gene expression changes.

Main Results:

  • Cell cycle gene expression, including Cyclin-dependent kinases (CDKs) and cyclins, peaked at passage 6 and declined by passage 8.
  • Pathway analysis consistently highlighted the cell cycle as a significant factor across all passages.

Conclusions:

  • Cell cycle gene expression dynamics can serve as a biomarker for TMSC state.
  • Passage 6 represents a potential window for inducing differentiation in TMSCs to maximize their therapeutic potential.
