A new electrospray-assisted laser desorption/ionization with gas transportation (ELDI-GT) method simplifies mass spectrometry imaging (MSI) sample preparation. This technique enables high-throughput analysis and accurate visualization of molecules without tedious sample processing.
Area of Science:
- Analytical Chemistry
- Spectroscopy
- Materials Science
Background:
- Mass spectrometry (MS) is crucial for label-free, multi-analyte measurements.
- Atmospheric pressure MS imaging (MSI) typically involves complex sample preparation.
- High-throughput analysis demands simplified ionization methods with minimal sample handling.
Purpose of the Study:
- To develop a novel ion source for mass spectrometry imaging (MSI) that eliminates the need for extensive sample preparation.
- To introduce a simplified, high-throughput ionization technique for MSI applications.
- To investigate the efficiency of gas transportation (GT) in enhancing ion signal intensity for MSI.
Main Methods:
- Development of an electrospray-assisted laser desorption/ionization with gas transportation (ELDI-GT) ion source.
- Utilizing a heated tube for nitrogen gas transport of the sample to the electrospray.
- Optimization of flow rate and temperature for ion signal enhancement.
- MSI analysis of caffeine crystals at a 50 µm pixel size.
Main Results:
- The ELDI-GT method requires no sample thinning, curing, planarization, or matrix addition.
- Ion signal intensity increased 11-fold by optimizing gas transport conditions (flow rate and temperature).
- Accurate visualization and localization of caffeine crystals were achieved at a 50 µm pixel size using MSI.
Conclusions:
- ELDI-GT offers a streamlined approach to MSI, significantly reducing sample preparation time and complexity.
- The developed ion source enables efficient gas transportation and enhanced ion signal for high-throughput MSI.
- ELDI-GT facilitates precise molecular localization in MSI without compromising sample integrity or requiring additional reagents.
