Area of Science:

  • Oncology
  • Molecular Biology
  • Epigenetics

Background:

  • Prostate cancer prognosis varies and is currently assessed using Gleason score and PSA.
  • Molecular biology advances enable identification of novel biomarkers for improved prostate cancer classification.
  • DNA methylation, an epigenetic process, is a promising area of research in cancer biology.

Purpose of the Study:

  • To review the role of DNA methylation in prostate cancer.
  • To summarize genes involved in prostate cancer carcinogenesis via methylation.
  • To discuss the diagnostic and prognostic implications of DNA methylation in prostate cancer.

Main Methods:

  • Literature review of scientific articles on DNA methylation in prostate cancer.
  • Focus on advancements in DNA methylation analysis techniques, including digital PCR.
  • Emphasis on the application of these techniques to circulating tumor DNA.

Main Results:

  • Identified various genes with methylation patterns linked to prostate cancer development.
  • Summarized the clinical implications of these methylation changes for diagnosis and prognosis.
  • Highlighted the potential of methylation monitoring for predicting treatment response.

Conclusions:

  • DNA methylation is a significant factor in prostate cancer carcinogenesis with diagnostic and prognostic value.
  • Monitoring DNA methylation, particularly in circulating tumor DNA, shows promise for treatment response prediction.
  • Current evidence is largely from retrospective studies, necessitating prospective research for validation.
