Summary

Circulating cytokines, including IL-8 and interferon-gamma (IFN-γ), can predict treatment response and prognosis in esophageal squamous cell carcinoma (ESCC) patients undergoing chemoradiotherapy (CRT). Low IL-8 and IL-1β levels indicate a favorable response to CRT.

Background:

  • Esophageal squamous cell carcinoma (ESCC) presents a significant challenge due to its poor prognosis.
  • Chemoradiotherapy (CRT) is a primary treatment modality for ESCC.
  • Identifying reliable predictors of treatment response and prognosis is crucial for optimizing patient care.

Purpose of the Study:

  • To investigate the potential of circulating cytokines as biomarkers for predicting treatment response and prognosis in ESCC patients receiving CRT.
  • To correlate pre-treatment and post-treatment cytokine levels with clinical outcomes.

Main Methods:

  • Serum samples from 36 ESCC patients were analyzed for 12 cytokines using multiplex immunofluorescence.
  • Statistical analyses, including Wilcoxon unpaired rank tests and Kaplan-Meier survival analysis, were employed.
  • Progression-free survival was monitored via imaging and follow-ups.

Main Results:

  • Elevated pre-treatment IL-8 levels were associated with lymphoid metastases.
  • Lower baseline IL-8 and IL-1β levels correlated with a partial response to CRT.

Conclusions:

  • Low levels of IL-8 and IL-1β suggest a favorable response to CRT.
  • Elevated levels of these cytokines may indicate a poorer clinical outcome in ESCC patients.
