Background:

Potato (Solanum tuberosum) is a vital global food crop, but functional genomics lags due to the absence of a suitable experimental model.
Existing potato research is hindered by limitations in genetic manipulation and a lack of standardized experimental lines.

Purpose of the Study:

To develop and characterize a novel diploid potato line, 'Jan', optimized for functional genomics and gene editing applications.
To create miniaturized 'mini-Jan' variants for enhanced high-throughput research in controlled environments.

Main Methods:

Developed a highly homozygous diploid potato line ('Jan') through seven generations of self-pollination.
Generated a chromosome-scale genome assembly for 'Jan', including gene annotation and syntelog identification.
Utilized CRISPR/Cas9-mediated mutagenesis to create 'mini-Jan' lines with dwarf stature by targeting growth-related genes (Dwarf and Erecta).

'Jan' exhibits high homozygosity, vigor, fertility, and excellent regeneration and transformation efficiencies.
