Area of Science:

  • Wound Healing Research
  • Cellular Biology
  • Biomedical Engineering

Background:

  • Chronic wounds present a significant clinical challenge due to impaired healing.
  • Pyroptosis, an inflammatory form of programmed cell death, is implicated in the pathogenesis of chronic wounds.
  • Hyperbaric oxygen (HBO) therapy is a potential treatment modality for promoting tissue repair.

Purpose of the Study:

  • To establish a rat model of chronic wounds.
  • To investigate the effects of HBO on chronic wound repair and pyroptosis.
  • To explore the role of pyroptosis in chronic wound pathogenesis and the mechanism of HBO therapy.

Main Methods:

  • Establishment of a rat model for chronic wounds.
  • Intervention with HBO and a Caspase-1 inhibitor (VX-765).

  • Assessment of wound healing, histopathology (HE staining), ultrastructure (TEM), protein and gene expression (Western blot, qRT-PCR), and inflammatory cytokine levels (ELISA).
  • Main Results:

    • HBO treatment significantly accelerated wound healing and improved pathological outcomes compared to the control group.
    • HBO therapy upregulated vascular endothelial growth factor A (VEGF-A) expression.
    • HBO therapy downregulated the expression of pyroptosis-related molecules, including NLRP3, Caspase-1, GSDMD, IL-1beta, and IL-18.

    Conclusions:

    • HBO therapy effectively promotes the healing of chronic wounds in a rat model.
    • The therapeutic effect of HBO may be mediated through the regulation of pyroptosis.
    • Targeting pyroptosis presents a potential therapeutic strategy for chronic wound management.
