Summary

PROK2 treatment mitigated oxidative stress and inflammation in testicular torsion/detorsion (TTD) rat models. This peptide therapy improved antioxidant capacity, reduced inflammatory markers, and enhanced testosterone levels, suggesting therapeutic potential for TTD-induced testicular injury.

Area of Science:

  • Biomedical research
  • Reproductive biology
  • Pharmacology

Background:

  • Testicular torsion/detorsion (TTD) induces significant oxidative stress, inflammation, and tissue damage.
  • Understanding the molecular mechanisms underlying TTD injury is crucial for developing effective treatments.

Purpose of the Study:

  • To investigate the therapeutic potential of PROK2 in mitigating ischemia/reperfusion (I/R) injury in testicular tissue.
  • To evaluate the effects of PROK2 on oxidative stress, inflammation, and histopathological changes following TTD.

Main Methods:

  • A rat model of testicular torsion/detorsion was established.
  • PROK2 was administered intraperitoneally at different time points during the TTD procedure.
  • Biochemical markers (testosterone, TAS, TOS, TNF-α, IL-6), histopathology, and immunohistochemistry were assessed.

Main Results:

  • TTD induced decreased testosterone and total antioxidant capacity (TAS), with increased total oxidant capacity (TOS), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6).
  • PROK2 treatment significantly improved testosterone levels, enhanced TAS, reduced TOS, and decreased TNF-α and IL-6 levels.
  • Histopathological evaluation showed improved testicular tissue integrity and increased spermatogonia count in PROK2-treated groups.

Conclusions:

  • PROK2 demonstrates significant antioxidative and anti-inflammatory properties in the context of testicular I/R injury.
  • PROK2 may serve as a promising therapeutic agent for protecting testicular tissue against TTD-induced damage.
