Background:

  • Inflammation, particularly interleukin-1 beta (IL-1β), is implicated in depression pathophysiology.
  • Stress activates microglia via the NLRP3 inflammasome, leading to IL-1β production and depression.
  • Beta-hydroxybutyrate (BHB) can reduce depression by inhibiting IL-1β, but its bioavailability is limited.

Purpose of the Study:

  • To investigate the potential of medium-chain triglycerides (MCTs) as a treatment for stress-induced depression.
  • To assess if MCTs can increase blood BHB levels and exert antidepressant effects.

Main Methods:

  • MCTs were administered orally to mice (social defeat model) and rats (chronic unpredictable stress model).
  • Blood BHB levels, immobility time in the forced swim test, hippocampal IL-1β levels, and spine density were analyzed.
