Summary

Adolescents with depression show altered brain connectivity, with decreased connectivity in some networks and increased connectivity in others. These brain network changes correlate with symptom severity, but the exact mechanisms require further study.

Background:

Purpose of the Study:

  • To investigate resting-state functional connectivity (FC) differences in adolescents with depressive episodes (DE) compared to healthy controls (HCs).
  • To explore the relationship between FC abnormalities and depressive symptom severity in adolescents.

Main Methods:

  • rs-fMRI scans were acquired from 55 adolescents with DE and 26 HCs.
  • Depressive symptoms were assessed using the Hamilton Rating Scale for Depression (HAMD-17).
  • Network-based statistical analyses compared FC between groups, using Yeo's seven-network scheme for seed regions.

Main Results:

  • Significant FC differences were found across seven networks in adolescents with DE.
  • Decreased FC was observed in the frontoparietal network (FPN) and sensorimotor network (SMN).
  • Increased FC was noted in the default mode network (DMN) and limbic network (LN); right superior parietal (SMN) and right precuneus (DMN) FC negatively correlated with HAMD-17 scores.

Conclusions:

  • Adolescents with DE exhibit widespread brain network abnormalities, including hypoactivity in external networks and hyperactivity in associative regions.
  • Observed FC changes suggest altered network interactions in adolescent depression.
  • Further research is needed to clarify the mechanisms linking these FC alterations to clinical symptoms.
