Background:

  • Colorectal cancer (CRC) is a leading cause of cancer mortality worldwide.
  • Early CRC detection is crucial for effective treatment but is hindered by asymptomatic or nonspecific early symptoms and invasive screening methods.
  • There is a critical need for improved, noninvasive screening tools for early CRC diagnosis.

Purpose of the Study:

  • To develop a label-free impedimetric immunosensor for the detection of colon cancer-secreted protein-2 (CCSP-2).
  • To create a cost-effective and noninvasive electrochemical tool for early CRC diagnosis and monitoring.
  • To assess the sensitivity, specificity, and detection limit of the developed CCSP-2 immunosensor.

  • Fabrication of a functionalized gold electrode modified with cysteine-modified recombinant protein G (RPGCys) to immobilize CCSP-2 antibodies.
  • Utilized bovine serum albumin (BSA) for sensor surface fouling prevention.
  • Employed Electrochemical Impedance Spectroscopy (EIS) and Cyclic Voltammetry (CV) to analyze electrochemical responses to CCSP-2 antigen-antibody binding.

  • Integration with other biomarker sensors could lead to advanced diagnostic and prognostic tools for CRC.
