Enzyme-Linked Immunosorbent Assay01:33

Enzyme-Linked Immunosorbent Assay

12.4K
In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or...

Summary
This summary is machine-generated.

The adiponectin and modified LDL (mLDL) complex protects blood vessels from mLDL-induced dysfunction. This study developed a method to detect the complex in human serum, finding higher levels in hemodialysis patients.

Area of Science:

  • Cardiovascular Biology
  • Adipose Tissue Biology
  • Vascular Physiology

Background:

  • Adiponectin is an anti-diabetic and anti-atherogenic protein.
  • Modified LDL (mLDL) contributes to atherosclerosis.
  • Adiponectin and mLDL form a complex influencing biological activity.

Purpose of the Study:

  • To investigate the protective role of the mLDL-adiponectin complex (MAC) in vascular tissue.
  • To develop and validate novel methods for quantifying MAC.
  • To compare MAC levels in hemodialysis patients versus healthy controls.

Main Methods:

  • Wire-myography on rat mesenteric artery to assess vasorelaxation.
  • Development of an ELISA using LOX-1 and anti-adiponectin antibody to measure MAC.
  • Comparison of serum MAC levels between hemodialysis patients and control subjects.

Main Results:

  • mLDL alone impaired vasorelaxation; co-administration with adiponectin (forming MAC) protected the endothelium.
  • In vitro, MAC detection increased with mLDL and adiponectin doses.
  • Hemodialysis patients had higher MAC levels and lower mLDL activity compared to controls.

Conclusions:

  • A novel ELISA successfully detected MAC in human serum.
  • MAC formation protects blood vessels from the harmful effects of oxidized LDL.
