Viral Mutations

32.1K
A mutation is a change in the sequence of bases of DNA or RNA in a genome. Some mutations occur during replication of the genome due to errors made by the polymerase enzymes that replicate DNA or RNA. Unlike DNA polymerase, RNA polymerase is prone to errors because it is not capable of “proofreading” its work. Viruses with RNA-based genomes, like HIV, therefore accrue mutations faster than viruses with DNA-based genomes. Because mutation and recombination provide the raw material...

Area of Science:

  • Virology
  • Immunology
  • Public Health

Background:

  • Dengue fever (DF), caused by Dengue virus (DENV), is a major global arboviral disease with 390 million annual cases.
  • The lack of specific antiviral treatments highlights the urgent need for effective Dengue vaccines.
  • Disease complexity and variant diversity necessitate robust vaccination strategies.

Purpose of the Study:

  • To review the current landscape of Dengue vaccine development.
  • To analyze various vaccine strategies, including attenuated, inactivated, subunit, and viral vector approaches.
  • To identify future research priorities and challenges in Dengue vaccine R&D.

Main Methods:

  • Comprehensive literature review of preclinical and clinical studies on Dengue vaccines.
  • Evaluation of vaccine candidates based on immunogenicity, safety, and efficacy.
  • Analysis of different vaccine platforms and their respective strengths and limitations.

Main Results:

  • Multiple Dengue vaccine strategies are under investigation, each with unique profiles.
  • Data from studies indicate varying levels of immunogenicity, safety, and efficacy across different vaccine types.
  • Key challenges identified include ensuring broad protection against DENV serotypes and achieving long-term immunity.

Conclusions:

  • Dengue vaccine development offers significant potential to reduce the global burden of DF.
  • Overcoming challenges in safety, efficacy, delivery, and accessibility is critical for successful implementation.
  • Advancements in vaccine technology are essential for enhanced control and prevention of Dengue.
