    Summary

    Orcein-positive granules of copper-binding protein (CBP) are key indicators in diagnosing specific adult liver diseases. These granules are absent in acute liver conditions but helpful in differentiating chronic biliary disorders and primary liver tumors.

    Background:

    • Copper-binding protein (CBP) and its orcein-positive granules are present in fetal and neonatal livers.
    • Physiologic levels of CBP in infants decrease to adult ranges by 3-6 months of age.
    • Distinguishing physiologic from pathologic CBP deposits is challenging in infants under 6 months.

    Purpose of the Study:

    • To investigate the patterns and incidence of orcein-positive CBP granules in liver biopsy specimens.
    • To assess the diagnostic utility of CBP granules in various liver diseases.
    • To correlate CBP granule presence with specific liver pathologies in adults and older children.

    Main Methods:

    • Analysis of 2,531 liver biopsy specimens from diverse patient populations.
    • Histochemical staining for orcein-positive granules of copper-binding protein (CBP).
    • Correlation of granule presence with diagnosed liver diseases, including Wilson's disease, chronic biliary disorders, cirrhosis, and primary liver tumors.
