Background:

Purpose of the Study:

  • To investigate the potential causal link between self-reported myopia and the risk of developing age-related cataract (ARC).
  • To provide genetic evidence supporting public health strategies for myopia and ARC.

Main Methods:

  • A two-sample Mendelian randomization study utilizing summary-level genetic data from the MR-Base platform.
  • 26 single-nucleotide polymorphisms (SNPs) for self-reported myopia served as instrumental variables.
  • Inverse-variance weighted (IVW) method as primary analysis, with weighted median, MR-Egger, and Maximum Likelihood as supplementary analyses. Sensitivity analyses (heterogeneity and pleiotropy assessment) were also conducted.

Main Results:

  • Mendelian randomization analysis indicated a significant causal effect of self-reported myopia on increased ARC risk (IVW: OR 10.657, P < 0.001).
  • Results were consistent across primary and supplementary analyses.
  • Sensitivity analyses confirmed no significant heterogeneity or horizontal pleiotropy, supporting the robustness of the findings.

Conclusions:

  • Self-reported myopia is causally associated with an increased risk of age-related cataract.
  • Findings support interventions aimed at preventing myopia onset and progression, and developing novel myopia therapies.
  • Further large-scale prospective studies are warranted to validate these genetic findings.
