M-Cdk Drives Transition Into Mitosis02:15

M-Cdk Drives Transition Into Mitosis

5.5K
Checkpoints throughout the cell cycle serve as safeguards and gatekeepers, allowing the cell cycle to progress in favorable conditions and slow or halt it in problematic ones. This regulation is known as the cell cycle control system.
Cyclin-dependent kinases, or Cdks, work in concert with cyclins to control cell cycle transitions. M-Cdk, a complex of Cdk1 bound to M cyclin, is a well-known example of this coordinated control that drives the transition from the G2 to the M phase.

The Retinoblastoma Gene01:20

The Retinoblastoma Gene

4.0K
Tumor suppressor genes are normal genes that can slow down cell division, repair DNA mistakes, or program the cells for apoptosis in case of irreparable damage. Hence, they play an essential role in preventing the proliferation of damaged cells.
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...

Summary

Cell Division Cycle-Associated (CDCA) genes are overexpressed in glioblastoma (GBM), acting as potential diagnostic biomarkers and therapeutic targets. Their elevated expression correlates with poor prognosis and tumor progression in GBM patients.

Area of Science:

  • Oncology
  • Molecular Biology
  • Genetics

Background:

  • Glioblastoma (GBM) is an aggressive brain tumor with limited treatment options.
  • Understanding GBM molecular drivers is crucial for developing new therapies.
  • Cell Division Cycle-Associated (CDCA) genes are understudied in GBM and may influence proliferation and treatment resistance.

Purpose of the Study:

  • To investigate the pathogenic, diagnostic, and therapeutic roles of CDCA genes in GBM.
  • To explore CDCA genes as potential biomarkers and therapeutic targets for GBM.

Main Methods:

  • RT-qPCR was used to analyze CDCA gene expression in GBM cell lines.
  • Bioinformatic analyses (TCGA, mutation, CNV, methylation) assessed prognostic and genetic roles.
  • Functional assays (gene knockdown, proliferation, migration) evaluated CDCA7 and CDCA8 in U87MG cells.

Main Results:

  • CDCA genes were significantly overexpressed in GBM cell lines and tumors compared to normal brain tissue.
  • CDCA gene expression demonstrated excellent diagnostic potential for GBM.
  • Higher CDCA gene expression correlated with poor prognosis and was linked to cancer-related pathways and immune modulation.
  • Knockdown of CDCA7 and CDCA8 reduced GBM cell proliferation, colony formation, and migration.

Conclusions:

  • CDCA genes are promising diagnostic biomarkers for GBM.
  • CDCA genes represent potential therapeutic targets for GBM treatment.
