Tumor Immunotherapy01:27

Tumor Immunotherapy

493
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Targeted Cancer Therapies

7.5K
The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...

Area of Science:

  • Immunology
  • Oncology
  • Biotechnology

Background:

  • Immune checkpoint inhibitors (ICIs) show promise for advanced melanoma but offer limited durable responses.
  • Stimulator of Interferon Genes (STING) agonists, like cyclic GMP-AMP (cGAMP), can enhance immune responses but face clinical translation challenges due to stability and off-target effects.

Purpose of the Study:

  • To evaluate the efficacy of Microbubble-assisted UltraSound-guided Immunotherapy of Cancer (MUSIC) in treating aggressive murine melanoma models.
  • To assess MUSIC's ability to synergize with ICIs and bridge innate and adaptive immunity.

Main Methods:

  • MUSIC utilizes microbubbles and ultrasound to selectively deliver cGAMP into antigen-presenting cells.
  • The study employed aggressive murine models of melanoma to test MUSIC's therapeutic potential.

Main Results:

  • MUSIC demonstrated curative effects in melanoma models, reducing tumor size and increasing overall survival.
  • MUSIC treatment synergized with ICIs, effectively bridging innate and adaptive immune responses.
  • The MUSIC strategy successfully activated STING and downstream proinflammatory pathways, leading to potent antitumor immunity.

Conclusions:

  • MUSIC is a potent immunotherapy for melanoma, showing curative potential in preclinical models.
  • This approach indicates MUSIC's promise as an adjuvant therapy for cancers currently treated with ICIs.
