Overview of Exosomes01:36

Overview of Exosomes

Exosomes are stable, lipid bilayer-enclosed vesicles capable of crossing biological barriers. They can carry a wide range of molecules required for intercellular communication. Once exosomes are released from the cell where they originated, they enter a recipient cell through various pathways such as fusion, receptor-mediated endocytosis, macropinocytosis, and phagocytosis.
Stahl et al. discovered exosomes in 1983, but the exosomes were initially considered waste products released from the...

Summary

Systemic microvesicles (MVs) from neutrophils and endothelial cells show promise as diagnostic biomarkers for breast cancer (BC). These findings support their potential use in minimally invasive screening tests for early BC detection and monitoring.

Area of Science:

  • Oncology
  • Biomarker Discovery
  • Flow Cytometry

Background:

  • Breast cancer (BC) affects millions globally, with prognosis dependent on accurate staging.
  • Systemic microvesicles (MVs) are linked to BC invasion, progression, and metastasis.
  • MVs offer potential for liquid biopsy in predicting treatment response and guiding individualized therapy.

Purpose of the Study:

  • To develop a minimally invasive diagnostic panel for breast cancer (BC) using circulating microvesicles (MVs).
  • To identify and validate MVs as complementary biomarkers for BC screening and follow-up.
  • To utilize machine learning for BC classification based on MV profiles.

Main Methods:

  • Quantification of circulating MVs in 100 BC patients and 48 healthy women via flow cytometry.
  • Analysis of MVs before and at 6, 12, and 24 months post-treatment in BC patients.
  • Application of machine learning algorithms to assess MV performance for BC identification and classification.

Main Results:

  • BC patients exhibited higher levels of neutrophil- and endothelial cell-derived MVs compared to controls pre-treatment.
  • Post-treatment, all MV populations decreased, with notable increases in leukocyte- and erythrocyte-derived MVs at 12 months.
  • Machine learning identified neutrophil- and endothelial cell-derived MVs as key indicators for BC diagnosis.

Conclusions:

  • Neutrophil- and endothelial cell-derived MVs are strong candidates for breast cancer (BC) diagnostic biomarkers.
  • These MVs show potential for use in screening tests for BC diagnosis.
  • The study highlights the utility of MVs in a minimally invasive approach to BC management.
