Thyroxine-induced changes in rat liver mitochondrial cytochromes.

M A Horrum, R B Tobin, R E Ecklund

    Molecular and Cellular Endocrinology

    Summary

Thyroxine injections increased rat liver mitochondrial cytochrome concentrations and altered their reduction levels during respiration. These changes may explain the increased respiration observed in hyperthyroid states.

    Area of Science:

    • Biochemistry
    • Cellular Respiration
    • Endocrinology

    Background:

    • Mitochondrial function is crucial for cellular energy production.
    • Thyroid hormones significantly impact metabolic rate and cellular processes.
    • Understanding cytochrome behavior under hyperthyroidism is key to metabolic research.

    Purpose of the Study:

    • To investigate the impact of thyroxine on rat liver mitochondrial cytochrome concentrations.
    • To determine how hyperthyroidism affects cytochrome reduction states during respiration.
    • To correlate changes in cytochrome levels and reduction with altered respiration rates.

    Main Methods:

    • Rats were injected with thyroxine for 10 days.
    • Spectrophotometry was used to measure cytochrome concentrations at specific wavelengths.

    Related Experiment Videos

  • Difference spectra in cytochrome c-depleted mitochondria assessed membrane-bound cytochromes.
  • Cytochrome reduction levels during state III and IV respiration were quantified.
  • Main Results:

    • Thyroxine increased concentrations of cytochromes b, c, total c, and a a3.
    • Cytochrome c1 concentration remained unchanged in both euthyroid and hyperthyroid rats.
    • Hyperthyroid mitochondria exhibited increased reduction of cytochromes b, c, c1, and total c during state III respiration.
    • Cytochromes c, c1, and total c showed increased reduction during state IV respiration.
    • Cytochrome b reduction decreased during state IV respiration in hyperthyroid mitochondria.

    Conclusions:

    • Hyperthyroidism alters mitochondrial cytochrome concentrations in rat liver.
    • Changes in both cytochrome levels and their reduction states are linked to increased respiration in hyperthyroidism.
    • These findings provide insights into the mechanisms underlying thyroid hormone-induced metabolic changes.
