Area of Science:

  • Cell Biology
  • Biochemistry
  • Membrane Biophysics

Background:

  • Lipid rafts are crucial for cell signaling and membrane protein function.
  • Their dynamic and transient nature poses challenges for in vivo study.
  • The lipid composition of the plasma membrane's outer leaflet dictates raft formation.

Purpose of the Study:

  • To develop and demonstrate a method for manipulating the lipid composition of the outer leaflet of living cell plasma membranes.
  • To enable the study of lipid raft characteristics and their impact on transmembrane protein activity.
  • To provide a protocol for lipid exchange using lipid-loaded cyclodextrins.

Main Methods:

  • Utilized lipid-loaded cyclodextrins for exogenous phospholipid exchange on the plasma membrane's outer leaflet.

  • Developed a protocol for preparing exchange media and treating attached mammalian cells.
  • Employed high-performance thin-layer chromatography (HP-TLC) to quantify lipid exchange efficiency.
  • Main Results:

    • Achieved nearly complete replacement of the outer leaflet with exogenous lipids.
    • Demonstrated that the lipid exchange method does not compromise cellular viability.
    • Successfully modified the plasma membrane lipid constitution of intact mammalian cells.

    Conclusions:

    • The described lipid exchange method offers a viable approach to study lipid rafts in living cells.
    • This technique facilitates research into the functional roles of specific lipids in membrane organization and protein activity.
    • The protocol provides a robust tool for investigating plasma membrane dynamics and function.
