This study presents a novel multi-dimensional, multi-mode, multi-column (3M) chromatography method for purifying alkaloids, a key source for drug discovery. The method successfully isolated 81 alkaloids, including 37 new compounds, from Menispermum dauricum.
Area of Science:
- Natural Product Chemistry
- Analytical Chemistry
- Pharmacognosy
Background:
- Natural products are vital in drug discovery, with alkaloids being particularly promising leads.
- Efficient purification of complex alkaloid mixtures is crucial for identifying new drug candidates.
Purpose of the Study:
- To develop and validate a novel multi-dimensional, multi-mode, multi-column (3M) preparative chromatography strategy for the effective purification of alkaloids.
- To demonstrate the resolving power and efficiency of the 3M strategy for alkaloid isolation from a complex natural source.
Main Methods:
- Development of a 3M preparative chromatography strategy integrating diverse separation modes, stationary phases, and mobile phase conditions for high orthogonality.
- Application of the 3M strategy to crude alkaloids from Menispermum dauricum DC for method validation and compound isolation.
- Characterization of purified compounds and assessment of the method's predictive capacity for larger-scale isolations.
Main Results:
- Successful purification of 81 alkaloids from 13.7 kg of M. dauricum using the 3M strategy.
- Identification of 37 novel alkaloid compounds, including 7 with previously unreported scaffolds.
- Demonstration of significant resolving power, with a prediction of isolating 445 compounds (>2 mg each) from 100 kg of M. dauricum.
Conclusions:
- The 3M preparative chromatography strategy is highly effective and rational for the efficient purification of alkaloids.
- This method significantly enhances the discovery of novel alkaloids and accelerates drug lead identification.
- The 3M strategy offers a powerful tool for natural product chemists and the pharmaceutical industry.

Chromatography: Introduction
The phase in which the compounds linger or on which the compounds adsorb is called the stationary phase, whereas the mobile phase is the solvent that carries the solutes to be analyzed. In traditional column chromatography, the mixture flows through the stationary phase, and the compounds partition between the stationary and mobile phases...

Band broadening refers to spreading solute bands as they travel through the column. This broadening can impact resolution. Plate height (H) represents the length required for one theoretical plate. A lower plate height corresponds to...
